Avvikelser i utvecklingen av hjärnbarken. Hit hör missbildningar till följd av onormal nervcellsutveckling eller celldöd, onormal nervcellsmigration, och onormal uppbyggnad av hjärnbarksstrukturen via nervutskott, synapsbildning eller nervcellsmognad. Förutom att mutationer direkt inverkar på utvecklingsprocesserna, så finns det ett antal medfödda ämnesomsättningstillstånd med sekundär inverkan, vilka också visar upp dessa missbildningar, som t ex peroxisomala sjukdomar, mitokondriesjukdomar och pyruvatomsättningsstörningar. De utgör en vanlig orsak till epilepsi och utvecklingsstörningar och ingår ofta i komplexa, medfödda anomalier.

Abnormalities in the development of the CEREBRAL CORTEX. These include malformations arising from abnormal neuronal and glial CELL PROLIFERATION or APOPTOSIS (Group I); abnormal neuronal migration (Group II); and abnormal establishment of cortical organization (Group III). Many INBORN METABOLIC BRAIN DISORDERS affecting CNS formation are often associated with cortical malformations. They are common causes of EPILEPSY and developmental delay.
